It reduces the risk of cardio vascular diseases, and blood vessels blockage.
Mechanism of action
Simvastatin is a prodrug and is hydrolyzed to its active ß-hydroxyacid form, simvastatin acid, after administration. Simvastatin is a specific inhibitor of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase, the enzyme that catalyzes the conversion of HMG-CoA to mevalonate, an early and rate limiting step in the biosynthetic pathway for cholesterol. In addition, simvastatin reduces VLDL and TG and increases HDL-C.
Contraindications
Contraindicated in patients with hypersensitivity, acute liver disease or unexplained persistent elevations of serum transaminases, during pregnancy and breastfeeding.
Do NOT use simvastatin if:
• you are allergic to any ingredient in simvastatin • you have liver problems or unexplained abnormal liver function tests • you are pregnant, may become pregnant, or are breast-feeding • you take cyclosporine, danazol, gemfibrozil, certain macrolide antibiotics (eg, clarithromycin, erythromycin), a hepatitis C virus (HCV) protease inhibitor (eg, boceprevir, telaprevir), an HIV protease inhibitor (eg, ritonavir), itraconazole, ketoconazole, mibefradil, nefazodone, posaconazole, or telithromycin • you take or have taken conivaptan within the past 7 days
Warnings and Precautions:
Caution should be exercised patients with history of liver disease. It may lead to rapid breakdown of skeletal muscle (rhabdomyolysis), severe infection, low blood pressure, major surgical trauma, uncontrolled seizures or severe metabolic, endocrine and electrolyte disorder. Avoid drinking alcohol otherwise it may result in liver failure. This medication should be used with extreme caution in children younger than 10 years old and in those who have not reached puberty. Discontinue treatment if there is marked or persistent increase in serum-aminotransferase concentrations.
Side Effects:
Central Nervous System- Headache, dizziness, sleeplessness, depression. Gastrointestinal- Nausea, flatulence, distortion of the sense of taste, abdominal pain, diarrhea, constipation. Eye- Lens opacities, blurring of vision. Metabolic- Serum transaminases and CPK elevations. Musculoskeletal- Muscle pain, muscle weakness and Immune-Mediated Necrotizing Myopathy (IMNM). Miscellaneous- Heartburn, hypersensitivity, sexual dysfunction, and upper respiratory symptoms.
How to use:
Use as directed by your doctor. Check the label on the medicine for exact dosing instructions.
• Take this drug by mouth with or without food, preferably in the evening, unless directed otherwise by your doctor. • Eating grapefruit or drinking grapefruit juice may increase the amount of simvastatin in your blood, which may increase your risk for serious side effects. The risk may be greater with large amounts of grapefruit or grapefruit juice. Avoid large amounts of grapefruit or grapefruit juice (eg, more than 1 quart daily). Talk with your doctor or pharmacist if you have questions about including grapefruit or grapefruit juice in your diet while you are taking simvastatin. • Continue to take simvastatin even if you feel well. Do not miss any doses.
Storage Conditions:
Oral: Store at 15-30°C. Store it in an airtight container and keep away from children. Protect from heat, light and moisture.
